The first step is to start up Adobe Illustrator and create a new file (in this case a size of 1920x1080 pt or px.
Then place the sketch on a separate Illustrator layer and lower its opacity so you can draw the flat illustration underneath it.
I would always recommend starting by creating the base shapes, then moving into creating the details (face of the character, separation lines), and only adding colors to your flat design at the end.
The last step is to create a few shadows to give the design a bit of dimension.
