Single Dose Administration, And The Influence Of The Timing Of The Booster Dose On Immunogenicity and Efficacy Of ChAdOx1 nCoV-19 (AZD1222) Vaccine
papers.ssrn.com/sol3/papers.c...
Background
Approved for emergency use by MHRA
Two standard doses, 4 to 12 weeks
Pooled analysis of trials
Exploratory analyses, immunogenicity and efficacy of extended intervals
Priming and booster doses
Data after one dose
Methods
Phase III efficacy trials in the United Kingdom and Brazil
Phase I/II clinical trials in the UK and South Africa
Data cut-off, 7th December 2020
Over 18 years, randomised 1:1
Blinded independent endpoint review committee
Participants
N = 17,177 baseline seronegative participants
UK, n = 8,948
Brazil, n = 6,753
South Africa, n = 1,476
Infections after first dose
Positive infections, n = 619
Symptomatic infections, less than 14 days after dose, n = 332 (up by 201 from 131)
Primary analysis of overall vaccine efficacy
Including LD/SD and SD/SD groups, = 66.7% (57.4%, 74.0%)
Vaccine group
Hospitalisations = 0
Control group
Hospitalisations = 15
Hospitalisations after a single dose
Less than 22 days
2 in vaccination group, 7 in control group
More than 22 after first dose
0 in the vaccination group, 6 in the control group
Vaccine efficacy after a single standard dose
From day 22 to day 90 = 76%
Protection did not wane during this initial 3-month period
Antibody levels were maintained during this 90-day period
Minimal waning by day 90
Efficacy with a longer prime-boost interval
VE = 82.4% at 12+ weeks
Efficacy with less than 6 week, prime-boost interval
VE = 54.9%
Observations supported by immunogenicity data
Binding antibody responses more than 2-fold higher after an interval of 12 weeks or more
Compared with and interval of less than 6 weeks
(18-55 years)
Interpretation
UK single dose strategy, booster at 3 months
is an effective strategy for reducing disease,
and may be the optimal for rollout of a pandemic vaccine when supplies are limited in the short term
It is the dosing interval and not the dosing level which has a great impact on the efficacy of the vaccine
Longer prime-boost intervals done with other vaccines such as influenza, Ebola and malaria.
Professor Andrew Pollard, Chief Investigator of the Oxford Vaccine Trial
These new data provide an important verification of the interim data that was used by more than 25 regulators including the MHRA and EMA to grant the vaccine emergency use authorisation
It also supports the policy recommendation made by the Joint Committee on Vaccination and Immunisation (JCVI)
for a 12-week prime-boost interval, as they look for the optimal approach to roll out, and reassures us that people are protected from 22 days after a single dose of the vaccine
Vaccinated people less contagious
Analyses of PCR positive swabs in UK
Vaccine may have substantial reducing effect on transmission of the virus
67% reduction in positive swabs among those vaccinated

This description of the most vulnerable accounts for the vast majority of Covid deaths and the median age of those who’ve died of COVID-19 is slightly older than the median age of those who died of all other causes. However, the majority even of this elderly group survive infection. Overall, the lethality of the virus is now known to be very close to typical seasonal influenza. Notably, in relation to risks to the working population, the lethality of the virus in those aged 60 and younger is actually less than seasonal flu.” Dr Mike Yeadon over 35 years experience in drug development for respiratory conditions. Going to trust him over corrupt politician and rushed vaccines with no longterm efficacy. You need to listen to people at the top of their field with nothing to gain. • Hi Dr John, and thank you for the updates. This far I have only one thing I had to disagree on. When it comes to agriculture, please do not think it is as simple as reducing animal, and animal contacts. We need animals to keep the soil happy, ( and us) They are part of the ecological system. To be able to eat good nutritional veggies we need good soil. Animals must be treated well since we are all in a symbiosis. Wild animals are wild and need their habitats. If we as humans could treat other living beings with respect - we would not have this problems. Stressed animals get sick, healthy animals is no risk whatsoever. A big hug to you for keeping us sound together with my animals.

    @maritatauni7351@maritatauni73513 жыл бұрын
    • Eating animals is unsustainable and animal agriculture makes people a lot of money for exploiting and killing animals. Where money is involved, ethical treatment will not exist. Factory farming is the result and it is a blight on humanity and has led to zoonotic viral infections. With the high demand for meat, factory farms are the only way to meet that demand. Factory farming causes great environmental damage as well as contributing to climate change. It is cruel, heartless, and unethical. It is bad for your health. Plant based diets are better for your health, better for the environment, better for the climate, and if you care at all about the animals of the planet, other than your precious dog or cat, you would realize that it is the only way to eat that is not unspeakably horrendous. Dr. Campbell is presenting the science as always, which confirms the many, many benefits of adopting a mainly plant based diet.

You then go on-line and scroll through the selection of venues currently available in your area and select the one where you hope to get the vaccine you prefer. Once you are of qualifying age you don't ever lose your elligibility (ie if you don't turn up at the specified time or you decide to say no thanks to the vaccine on offer) and you can always rebook later on.

If the data is only for 18-55 years olds, and its generally people OVER 60 who are most likely to be hospitalised, it doesnt really answer the question about its efficacy for the main group who are likely to end up in hospital (ie, people over 55)

But they have side effects. Nobody who takes upto 48mg ivermectin had side effects So it is ok to take 48mg ivermectin .it is the best effective.. But fda recommends 24mg over 16years old. In the third and final phase, 20 thousand people took part in the study. Three quarters of them received the vaccine, the other quarter a placebo. Based on the group that received the vaccine, it was found to be very effective. After two injections, the average effectiveness is 91.6 percent. In people aged 60 and older, the effectiveness was 91.8 percent. Also striking: after the first dose of the drug, the effectiveness already appears to be 73.6 percent. These results would be better than those of AstraZeneca and the Dutch pharmaceutical company Janssen. 👍👍👍

Remember, there's too many of us who can't or won't take these vaccines (40% in North America) including health workers and the 1%, and those individuals will not allow themselves to become second class citizens.
