Plant Extraction Methods - Decoction and Maceration | JPTV
Hey! Are you even curious about how MX3 gets "xanthone" from mangosteen or how Myra-E extracts "d-alpha tocopherol (vitamin E)" from soybean oil? Typically, we see in the advertisements of whitening, antiaging, and food supplement products the terms like "ascorbic acid (vitamin C)" in Ceelin, "resveratrol" in Slenda, or "glutathione" in Vitapack Megawhite. These terms are names of bioactive compounds that exhibit tons of health benefits like antioxidant, anti-inflammatory, antibacterial, antifungal, antidiabetic, antiviral, and even anti-cancer properties.
Pharmaceutical scientists extract these bioactive compounds from different natural sources for medicinal purposes using different extraction methods. If you are curious about how they do it, watch this vlog. #JPTV #NeverStopLearning

This is a very comprehensive guide
Thank you, sir! This has helped me in understanding better the procedures I need to do for my thesis.💚
You're writing a thesis but you need to go on KZhead to figure out how to run a rotovap?

Thank you for sharing your knowledge po ☺
Thanks doc. Learned a lot
Nice content lalo na for students. Very helpful 👏🏻👏🏻👏🏻👏🏻
2:43 please.. have a question.. what type of solvent you have used for maceration ? I have another question : for ex if we use ethanol-water as solvent, after the extraction we eleminate the ethanol first using ratary-evaporator,but how we eliminate water fraction ? Thanks brother for sharing this video !!
Hi, Sir Doc, I find your video very helpful in our on going project study, I just want to ask if we use boiling of the roots, only through centrifuge is applicable to get the final extract or we can use the rotovap? Thank you Sir! I also subscribed in your youtube channel.
your video is super helpful. thank you so much
Thank you ❤️
wow this is a great video thanks for this!!!
I would like to ask if there are other ways of extracting epicuticular wax as water resistant coating from leaves? We're just going to use it for our capstone project. Thank you!
in oil extraction, how would you compare maceration and cold pressed using a machine, which do you think is more potent?
This is very informative! Support ito, grabe!

I'm a 4th year pharmacy student and want to be involved with pharmaceutical science research. I've been thinking about taking a Masters in Pharmacy, but I'm second guessing because from what I see the type of research produced is usually assays of plant extracts which I kinda don't like. The type of research I want to do is along the lines of polymers, nanotech, etc. (i haven't quite figured it out yet). Do you have any advice for me?
Thnq for such a valuable video sir..... Sir can u pls resolve my quiery...if we want to prepare a plant extract (for any activity) for whom we dnt have any idea about its solubility then which solvent will we use?
Great video!
great work
Thanks doc!
great work sir , can you help me with a way to extract tannin from a plant ?
Hi sir. Are there any alternatives for rotary evaporator? Can you use simple heating to remove the solvent?
Thank you for sharing bro.....
You’re welcome!
Very nice sir... I have a question? Someone who don't have fuming hood then how can be evaporated the remaining solvent? As by keeping in open air it takes lot of time to evaporate... ur reply will be appreciated sir...
Excellent
Do you have published paper that i can cite you on this method? By the way, thank you for sharing this! Very much appreciated.
Hi Mira! Thank you so much for your comment. In my own study, I used this paper www.sciencedirect.com/science/article/abs/pii/S030881460900199X. But generally, this video is intended to show how to perform basic plant extraction and discuss what plant extract is. 😊
@@DoctorJPTV Alright, well noted. Thank you for sharing! Love your vid. Simple and easy to understand. 😊😇❤️
Sir I want to work on cannabis but don't know what to do because already some much is done
@DoctorJPTV, how can someone reach you? I’m trying to extract saponins from plant and I was considering using an alcohol like methanol or ethanol as the solvent while I use maceration process for the extraction I’m kinda confused if I should dry my plants first or just use the plant like that And another question, if you can help me with the procedure for maceration extraction I will be grateful Thank you doc!
hello! good am I am a 4th-year high school student I just wanna ask what is the best solvent to use for maceration for extracting the components in the plant and also good for the skin?
Geez Louise, and we all wonder why our skincare or medicinal extracts costs so much; well....Exhibit A, ladies and gentlemen!! I need to remember this AMAZING process, shown to us by the kind Doctor. the next time I'm shopping for extracts and whining that they "just cost too much" at $14.95 a bottle; am I right? Thank you, Doc, for opening my eyes and giving me some perspective!
hi Doc, do you have a related study about decoction method that we can use as a citation? we are currently working on a study but we are lack of citations
Thank you so much for uploading this video. ALLAH BLESS YOU ❤️❤️❤️
pls do a video proceeding to HPLC analysis :)
Sir Please suggest food wastes( plant or vegetables) for the extraction of polyphenols for my dissertation
Man I like this laboratory. Thanks for the video. This is the third time I am watching this. So after extraction how do you fortify your product with the extract?
By “fortify” do you mean purify? You need to do chromatic profiling, separation and purification techniques for that.
How to chose best solvent for Phthalates extraction? GC-MS analysis
hi sir! i'm currently in jhs and we're planning to extract wax from leaves. do u think this method is applicable?
Hey really nice video, so i wanted to extract anthocyanins from Jamun fruit using maceration with ethanol, do I need to dry the fruit or can I just simply put the fruit inside the solvent ?
Preferably dry the fruit to saturate the concentration of anthocyanin. Since anthocyanins are light sensitive make sure to dry it on a dark areas.
Good job
Hello sir I have ..I've kept coriander leaves for maceration (for antibacterial property against p.acne) .but only using water I did not use ethanol or methanol with the water ..is it possible to obtain the concentrated extract from it and if so wt should be the temperature maintained in the water bath and also the pressure
What is the shelf life of the "extracts" when performing extraction process? In my case, I consider Iyphilization/freeze drying the plant to achieve the same texture of green tea before boiling it. I wanted to store a large amount of dried leaves for future purposes and save it for decoction. Will it affect the shelf life?
he was doing Iyphilization/freeze drying after pretreatment in freezer.
Hi Sir, how many plant powder and solvent during maceration in this video? Thankyou
Good day! The plant powder that you used in the beginning of the video, was it store-bought or did you prepare the powder extract? Thank you!
We prepared the plant powder. We pulverize the leaves using a mechanical miller.
Im interested in plant extraction and recrystalization can you do a video for that
Thank u man💚💚
Hello doc! Pls answer this because I really need it for my team's defense How many extracts will be if I have 3kg of bougainvillea bracts using the maceration method?
so the vapour temperature is basically the solvent's boiling/evaporation point?
Hey doc how r u doing that was a great video. Can you please show us fraction distillation of essential oil to extract different components from it or any other technique. For example I want to seperate citrol and Other components

Sir I want to add natural vitamin E in my syrup formulation as my research work from vit e rich fruit or vegetable...so how to extract ?plz rply
Good day, I would like to ask if what plant extraction is applicable to acquire its epicuticular wax from it?
OMG thank you.
thank you
Thank you for sharing but I need more clarifications how can you help me?
This video is very insightful. May i ask if you don’t mind, when using the decoction method what is the reasoning behind freezing? And if necessary how will it be possible to obtain a liquid form of the extract instead of the powder? Thank you for any response and for this video.
Hello! Thank you for your comment. To answer your questions, 1st. the reason behind freeze drying is to remove the water content from your extract without deteriorating your bioactive compounds. 2nd. if you want to obtain a liquid form of your extract, it's either you can dissolve the powder in your treatment vehicle (solvent used treatment, depending on your experiment), or you can use the extract automatically after decoction and filtration, but you have to make it more concentrated. If you don't concentrate your extract, it will probably yield lesser bioactivity.
How we can concentrate the extrait
Hi Doc! We are currently working on a research where are going to extract Peroxidase enzyme from papaya to detect glucose in a urine through GOD-POD method, but we are struggling in finding a laboratory that will help us to extract the Peroxidase from Papaya so that we can have a analytical grade peroxidase that we can use for our research. Do you have laboratories that you can refer so that we can contact them?
Are you using Papaya leaves or Papaya fruit?
Thanks doc. What's the ratio of the extract to solvent?
Him Thanks for this very informative video. Can decoction be used for stem bark?
Yes 😊
@@DoctorJPTV Thank you. I'm currently doing a stem bark extraction using ethanol as my solvent. Do you know what procedure I could perform to powderized my liquid extract? or to make it a liquid gel? I'd love to hear from you. Thank you
hi ! how much of saluyot did you use in this procedure?
Hi doc! What did you use po for filtration? Which is better po to use for filtration, cartridge of UF membrane or qualitative filter paper?
Hi sir, may I ask how many grams are in the vial bottle after rotary evaporation. Thank you
Hi! your video helped me so much. But can I ask a few questions to clarify? 1. What is the best solvent to use for decoction method for extracting phenolic content? 2. What is the best solvent to use for decoction method for extracting flavonoid content? Your response would be very much appreciated because I need this for my research methodology. Thanks!!
1 and 2. You can only use water in decoction method. You’ll be extracting flavonoids and phenolic compounds that are soluble in water only.
I wanna ask how much the ratio of plant sample and solvent do you use. If i use 5g of dried plant how much of ethanol do i need to dissolve the sample?
What type of solvent are you using?
what solvent did you use po for the extraction? i am working on my research paper similar with this methodology however i don’t have enough knowledge about it since i am only shs student and i don’t have consultant as well bcs of this pandemic.
I used 99% analytical grade ethanol. If you need some help on your research paper, feel free to email me at doctorjohnpaulmedia@gmail.com.
Hehe! Kawa ka mo ng mga video about make shift lab equipment since hindi naman lahat may boiler at vacuum ek ek. Para sa mga mahilig mag experiment pero wala pambili ng equipment. Hehe
Hi. This is really informative, can I ask what are the exact volume / measurement of the used materials here? What is the ratio of the plant to solvent? Will there be a difference in the result if the amount of extracted plants are bigger? Thank you.
There’s no need to follow any ratio. As long as you have totally submerged your plant sample in your solvent then you’re good to go. After all, will remove the solvent when you run your mixture in a rotary evaporator.
@@DoctorJPTV thank you! We're planning to make different concentrations for the plant extract to determine which is more effective as antibacterial. How do we measure or make different concentrations of the extract?
@@kaeyaalberich4427 you just have to weigh certain amount of your extract and dissolve it in a certain volume of solvent medium. Example: 10mg of plant extract dissolve in 10mL water will make 1 mg/mL plant extract.
Hi. What HPLC machine will be good enough to read Thc content pritty exakt?
I have a stupid question but please help, i want to extract nutrients from Seaweed to use it as fertilizer does boiling water denature or destroy any useful amino acids or plant hormones like auxins or cytokinin etc?
was hoping u would make the hplc ms/ms portion as well (:
I always heard extractions where done with alcohol, butane, propane, and co2 Infusions where done with water, oil, viniger or a low proof alcohol like wine ?
Hello Dr. JP hope all is well, I was wondering if there is a less destructive method of extraction there. All of this heating and drying surely would destroy much of the plant original compounds leaving us is a big chunk of oxidized and a lower grad less therapeutic extract... Would love to hear your answer, thank you.
Personally, I agree with you. However, I don’t thinking you’re making too much alterations or destructions to the bioactive compounds as long as you are in the limits. Hence, it is important to really know the your target bioactive compounds, such as their boiling point, melting point, polarity, solubility, etc.
@@DoctorJPTV thank you for your timely response I am definitely learning something new with your video. But my question remains unanswered if you may. Do we have an alternative to all of this procedure? I am trying to be as puritan as I can in my preperations and know in my gut that there must be a friendlier less destructive alternative. Thank you again 💖
You can try decoction method. However, using this method you can only extract water soluble bioactive compounds.
@@DoctorJPTV ok got you Dr. 👌 a LOT of lerning still lays ahead for me 🙏
Hi doc, can you make content about extracting polyglutamic acid from soybean and turning into a powder?
Is this method of wet extraction of inulin from sisal bole???
Hello! Thank you for this informative video. I have 2 questions to which I hope to have an answer to, it would be very much appreciated. 1- In our lab, we don't have a freeze dryer (lyophilisateur), and in a lot of references, i read that they used a drying oven at 40°C for 48h to evaporate to water in the extract. It is okay? Will it not affect my results too much? 2- For your alcohol extract, after using the rotavapor, the extract was liquid meaning there was still a little bit of solvent in it no? if i'm right, how will you then calculate the concentration if you don't know the initial concentration of your extract? Thank you!
1. Yes, you can do that. 2. Yea, there will be some alcohol left. But you need to further evaporate this in a fumehood for a couple of days. Concentration of what? For what? It is automatic that the concentration of the extract you have collected is 100% pure crude extract. If you want to use it as a treatment for testing, then compute the specific concentration you need for the testing using weight of the extract/volume of solvent.
hi can u send me your basis? might as well use it in my reswarch thank u
Doc how many grams of plant powder was used for macerating?
Good day doc! I just want to ask if you have any home based alternative for extracting D-limonene from citrus peels?
I’m not sure but I think you need to perform steam distillation to get extract d-limonene.. try checking then form of D-limonene and look what steam distillation is. 😊
@@DoctorJPTV thank you doc! 🙏
How'd you identify what ratio of solute:solvent to use?
Which paper is used for filtration?
Good day, Doc. Doc, how long did you store your crude extracts in your fume hood for further evaporation? I mean how long does it take to make the crude extract from syrupsy to dried form, if possible? Thank you in advance, Doc.
Around 1-3 days.. it depends.. you’ll know it’s ready if you can’t smell the solvent from your extract and you can observe that it has dried up.
Thanks, Doc!
Hello sir. Good pm. I am a 4th year college conducting a thesis. I have few questions regarding in our study. we use Flower extracts to develop an alternative highlighter pen inks. now I want to know how to get the concentrations of the inks? I hope you can help me sir. thank you in advance 🙂
Good day Doc. Please which is a better polar solvent between aqueous and ethanol in extracting plants bioactive compounds against microbes?
Water is more polar than ethanol. If you want to extract bioactive compounds specifically those with antimicrobial properties, better check the literature of what Antimicrobial compounds present in your plant and identify in which solvent are they soluble.
I don't get it, ah why did u have to do the rotary evaporation(?) for plant extraction if the filtered solution on the first part is already the plant extract??
Hai, when you freeze dry. Is it ok to fill almost full or leave a small space in your centrifuge tube? Because the technician in the lab that I'm using freeze dryer would tell to fill just half. but I filled the same as in your video. Thanks.
We also fill half of our containers.
Good day sir, Is the extract from rotavap the final extract that is used for antimicrobial testing? Thank you so much and Im looking forward to hearing your response😊
Yes. But make sure that the solvent or alcohol used has completely evaporated.
@@DoctorJPTV Noted sir. If you don't mind, may I just ask one question po. What is the best alternative to use if our laboratory doesn't have rotavap? Thank you in advance sir☺️
Hi! If you have no plan on retrieving your extraction solvent (alcohol), you can just use a typical water bath. Maintain a certain temperature, use the boiling point of your solvent or somewhere close to that temperature. Then continuously stir your extract during the evaporation process.
@@DoctorJPTV Thank you sir. God bless you always❣️
Good afternoon, Doc. Can we remove the water using rotary evaporator? We have used 80% ethanol and methanol in the maceration process. However, we are planning to further concentrate the crude extract. What is the alternative way po? Thank you, Doc!
May I know how can I determine the chemical properties of a certain plant?
hello doc, your video is a big help to me, May I ask, what is the recommended ratio of solid to solvent when performing maceration. thank you so much
There is no recommended ratio of solid to solvent when performing maceration because the end step is totally evaporating the solvent. So, as long as you have completely submerged your plant sample in your solvent then it’s good.
How to extract Bisphenol A (phytoremediated) from plants... Plz help
Awesome video shall I know the best method for extracting Ginseng herb? can we use the same method? please advise and thanking in advance
It really depends on your target bioactive compounds present in Ginseng herb.
thanks. one query is if you have prepared ethanol extract then how to incorporate it in feed during feed formulation.?
Before introducing you treatment (extract) to animal models you should make sure that all of the extraction solvent (ethanol) has completely evaporated. Usually, you can administer your treatment using the common drug delivery methods like oral, intravenous, intraperitoneal, etc. You can also add your treatment to the feed formulation but it will depend on what kind of feeds you have and its nature (dry/wet). Moreover, adding your treatment to the feed formulation might lead to inaccuracies in the volume or concentration of the drug (extract). So, you have to take note of that too. Like for example, if you want to treat your animal model with 200uL of 10 mg/kg extract, you plan how you can administer this accurately to all of your replicates to avoid biases and such.
what solvent did you use in maceration of the plant? :)
I used different extraction solvents. I used ethanol, methanol, DCM, hexane, toluene, etc.
How to extract any mineral from the plant? Thanks.
Hi! May I ask what is the proper measurement of your solvent and your grind products?
Hi! For this experiment, I used 95% ethanol and my plant material was powdered Corchorus olitorius leaves.
Bula .... Do you also extract parapheremone ?
Hello! Sir, may I ask about apple maceration?. We're planning on getting its extract, but we're not sure if we still need to dry them or it's fine to just cut it into small pieces?. Thank you!
Just cut them in smaller pieces. If you have a dehydrator you can also do it then pulverized the fruit prior to maceration.
@@DoctorJPTV noted, thank you so much!!
Hi, did you get the plant powder by using mortar and pestle?
We freeze-dried our plant sample and pulverized it using a milling machine.
